Fair to say that we all assume that aging is inevitable. In reality however, there is no biological law that says we must age. Over the years we’ve seen a variety of theories proposed to explain why we age including the accumulation of damage to our DNA, the damaging effects of chemicals called “free radicals, changes in the function of our mitochondria, and so many others.

Our guest today, Dr. David Sinclair, believes that aging is related to a breakdown of information. Specifically, he describes how, with time, our epigenome accumulates changes that have powerful downstream effects on the way our DNA functions. Reducing these changes to the epigenome is achievable and in fact, even taking it further, his research now reveals that the epigenome can be reprogrammed back to a youthful state.

David A. Sinclair, PhD, AO is Professor of Genetics at Harvard Medical School, and is the author of Lifespan — Why We Age and Why We Don’t Have To. He is the Founding Director of the Paul F. Glenn Center for the Biological Mechanisms of Aging at Harvard. One of the leading innovators of his generation, he is listed by TIME magazine as one of the “100 most influential people in the world” (2014) and top 50 most important people in healthcare (2018). He is a board member of the American Federation for Aging Research, a Founding Editor of the journal Aging, and has received more than 35 awards for his research on resveratrol, NAD, and reprogramming to reverse aging, which have been widely hailed as major scientific breakthroughs and are topics we discuss in our time together.

In 2018, Dr. Sinclair became an Officer of the Order of Australia, the equivalent of a knighthood, for his work on national security matters and human longevity. Dr. Sinclair and his work have been featured on 60 Minutes, Today, The Wall Street Journal, The New York Times, Fortune, and Newsweek, among others.

The gel, which is known as RM191A, is believed to be a new class of anti-inflammatory.

It’s a copper-based compound which is applied directly to the skin as a topical.

Summary: Scientists have long marveled at the rejuvenating effects of heterochronic parabiosis. When you mix the blood of a young mouse and an old mouse by joining their circulatory systems, the older animal recovers some features of youth, while the young animal becomes functionally older. While many have assumed that these effects were driven by the infusion of pro-youth factors from the young parabiont into the older one, an alternative “Dilution Solution” hypothesis is possible: that the young blood is instead diluting pro-aging factors from the old animal’s blood, as well as allowing the young animal’s livers and kidneys to filter out metabolic toxins through the young animals’ livers and kidneys.


In heterochronic parabiosis, joining the circulatory systems of young and old mice causes the older animal to recover some features of youth. The effect has been widely assumed to be driven by pro-youth factors in younger blood, but an alternative hypothesis is possible: that the procedure is instead diluting pro-aging factors in the older partner.

Gene therapies are opening up possibilities that were once reserved for science fiction.

At Harvard University, Professor of Genetics David Sinclair says he believes it’s possible to unlock the fountain of youth, and gene therapy is the key.

Sinclair spent two years trying to correct the vision of a mouse using gene therapy, and finally succeeded in doing it.

Although these discoveries are encouraging, Sinclair cautions that people set their expectations realistically.

“Many people are eager to use the research for their own health benefit,” he said. “But I’m hoping that the public will realize that it does take a long time and we can’t just jump from a mouse to a human tomorrow.”

Aging researcher Dr. Nir Barzilai is studying clinical trials that use the diabetes drug metformin to directly target aging.
